Title of the talk:Bacillus Spore Probiotics: Enhancing Diarrhea Treatment in Children while Reducing Antibiotic Dependence by Regulating Immune Responses and Restoring Gut Microbiota
Abstract
Persistent diarrhea poses a significant threat to children’s health in developing nations, presenting as a severe gastrointestinal illness with elevated risks of mortality. We conducted a double-blind, controlled clinical trial to assess the impact of high-dose liquid Bacillus spore probiotics on treatment outcomes. The trial featured two formulations: LiveSpo CLAUSY (containing B. clausii) at >2 billion CFU/5 mL ampoule, and LiveSpo DIA30 (comprising B. subtilis, B. clausii, and B. coagulans) at >5 billion CFU/5 mL. Administered at 4–6 ampoules daily, these probiotics significantly expedited recovery, shortening the duration of illness by 2-3 days (p < 0.05) and demonstrating 1.5–2.4 folds greater efficacy in alleviating diarrhea symptoms, including high frequency of bowel movement of ≥ 3 stools a day, presence of fecal mucus, and diapered infant stool scale types 4-5B. Remarkably, the probiotic intervention groups reduced a 25% in antibiotic use compared to the Control group, attributed to 2-day shorten antibiotic treatment duration (p < 0.05). By day 5 of treatment, levels of pro-inflammatory cytokines TNF-α, IL-17, and IL-23 decreased by 3.24% – 29.76% (p < 0.05), alongside a significant drop (p < 0.05) in excreted IgA in stool. 16S rRNA gut metagenomic analysis revealed increased densities of Bacillota phylum and beneficial Lactobacillaceae and Streptococaceae families, coupled with decreased density of Proteobacteria phylum and harmful Enterobacteriaceae family in the probiotic intervention groups. In summary, LiveSpo CLAUSY and LiveSpo DIA30 emerge as potent agents for pediatric diarrhea management, offering symptomatic relief, immunomodulation, and gut microbiota restoration.

Nguyen Thi Van Anh attained her PhD at the age of 28 from Tohoku University, Japan and has accumulated 17 years of experience in lecturing at Vietnam National University, Hanoi University of Science. Presently, she serves as a scientific consultant for pharmaceutical and biotechnological companies, including ANABIO R&D and LiveSpo Pharma, dedicated to the mission of achieving “a future without antibiotics.” With a track record of 35 publications in high-ranking journals and 5 patents to her name, she is recognized as an experienced professional in the Bacillus spore probiotics field.
