Title of the talk: Preclinical Models of Infectious Diseases for the Development of Novel Drugs and Vaccine to Combat Antibiotic-Resistant Pathogens

Abstract

Funded by the United States National Institutes of Health, Food and Drug Administration, and more than a dozen companies, the Diep lab at UCSF performed preclinical efficacy testing of various antimicrobial agents, antibacterial antibodies, and vaccine candidates against genetically diverse and drug-resistant strains of Staphylococcus aureus and Pseudomonas aeruginosa in our rabbit infection models.  I will highlight preclinical efficacy data from our rabbit models of non-ventilated pneumonia, ventilator-associated pneumonia, and hyperdynamic septic shock that advanced into clinical trials several novel anti-bacterial drug and vaccine candidates to combat the major antibiotic-resistant pathogens.

 

Biography

Dr. Diep is a Professor at the University of California, San Francisco.  Dr. Diep completed his BA and PhD degrees at UC Berkeley and postdoctoral training at UCSF.  Dr. Diep has a long-standing research program in the clinical and molecular epidemiology and pathogenesis of drug-resistant infections (70+ publications, 11,000+ citations). His group was among the first to characterize the emerging epidemics caused by the community-associated MRSA clone USA300. He directed NIAID R01-sponsored development of a rabbit non-ventilated pneumonia model that demonstrated mechanism by which Panton-Valentine leukocidin (PVL) causes acute respiratory distress syndrome, which was published in PNAS in 2010, and that neutralization of PVL and alpha-hemolysin by human antibodies conferred protection, which was published in Science Translational Medicine in 2016.  Over the past 8 years, Dr. Diep received $7M+ for preclinical animal studies to evaluate novel candidate agents targeting Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae, and Acinetobacter baumannii, which was funded by NIH/NIAID, Food and Drug Administration, Department of Defense DARPA, CARB-X, and industry partners, including Achaogen, Arsanis, AstraZeneca/MedImmune, ContraFect, Exbaq, Integrated Biotherapeutics, and Merck/Cubist.
